In the iPSC Generation Dox Inducible protocol, addition of Dox to the culture medium of transduced cells induces the reverse tetracycline transactivator （rtTA） to bind to the Tet response element and results in the transcriptional activation for each of the factors implicated in the reprogramming process. Conversely, Dox can be removed from the medium to suppress transcriptional activation of the reprogramming factors.
